A detailed in vitro multiplication system for rapid micropropagataion of the
Hypericum perforatum L. has been developed. Shoot tips and axillary buds
excised from this plant which were collected from Tartous explants and surface
disinifected then cultured on MS (Murashige and Skoog 1962) medium.
This research was conducated on Ceratonia siliqua L. in vitro to study the effect of
some growth regulators in both multiplication and rooting stage. And determine the best
concentrations wich lead the highest rate of multiplication (interms of number of shoots
and length), and the best rooting (interms of percentage of rooting, number of roots and
length).
This present study was conducted to develop a detailed in vitro propagation
system for the medicinal shrub Capparis spinosa L.
Single nodes with one bud and a small part of stem of 1-1.5 cm long were
used as initial explants which were collected f
rom a shrubs grown under field
conditions at Damascus suburb., (Doumar). Explants were surface-disinfected
by 70% Ethanol for 1 min., followed by immersion in Sodium Hypochlorite or
HgCl2 for different periods and concentrations with 1 drop of Tween 20 for 100
ml disinfectant solution, where after, they were placed onto MS basal medium
containing a combination of growth regulators at different concentrations (BA
at 4.44 or 8.88 μM) each with IBA 0.49 μM. Cultures were incubated in the
growth room at 23±1 c and light intensity of 3000 lux at the cultures level.
Multiplication rate of 25.17-fold from one explant was achieved every 4 weeks
on the optimal MS medium (MS+8.88μM BA+0.49μM IBA).
The described method has potential to produce large numbers of plantlets
within a short period of time to expand its cultivation for medicinal uses.
This study was conducted at the laboratories of Horticultural Department, Faculty
of Agriculture, Damascus University to determine the effect of using three levels of
sucrose (10, 30, 50 g /l) on chlorophyll content between healthy and vitrified pl
ants of
three varieties of Gypsophylla panicolata (Golantwo, Planshnet, Newlove). Results
showed superiority of treatment 10 g / l sucrose in reducing vitrification percent to
36.78% compared with 62.57% for control (30 g/l) and with significant differences
between varieties. Planshnet was the best variety in chlorophyll content either in healthy
or in vitrified plantlets. Moreover, the treatment with sucrose of 10 g/l caused significant
increment of chlorophyll content compared with other treatments. The high total
chlorophyll content was 17.46 micrograms/ ml in healthy plants compared with 14.26
micrograms / ml in vitrification plants for Planshnet at 10 g/l sucrose.
A successful and detailed in vitro propagation system for rapid
micropropagation of three apple rootstocks: MM ١٠٦, EM ٧ and M ٢٦ has
been developed. Shoot tips and axillary buds excised from field-grown trees
were used as explants, which were surface-disinfected with mercury bichloride,
or with solution of sodium or calcium hypochlorite followed by three rinses
with sterile distilled water.
Tissue culture process of date palm propagation via bud proliferation is
fully described. Adventitious plantlets were obtained from shoot tips. axillary
buds and the bottom of young leaves of five date palm cultivars (Sefri, Zahdi,
Rizzez, Khalas
and Hillaly).
