The objective of this research isto study the in vitro effect of some plant hormones (growth regulators) BAP,NAA, 2,4-D, and genotype on callus and bud formation from embryonic axes of two Soybean cultivar seeds (sb-44, sb-172). The embryonic axes were cultured on MS basal medium and MS supplemented with 1, 2, 3 mg/l BAP alone and in combination with 0.5 mg/l NAA.The cultures were maintained at 25 C°±1 with photoperiod of 16 hours light (2000-2500 lux) and 8 hours dark.The highest percentage92.5% and mean average 4.63 of callus formation were recorded on MS medium containing BAP (3 mg/l) and NAA (0.5 mg/l). The highest percentage 67.5% and average 3.38 of buds formation were obtained on MS medium supplemented 1 mg/l BAP and 0.5 mg/l NAA in cultivar sb-44. The percentage of callus formation increased, while the percentage of bud formation decreased with each increase in BAP concentration when used alone. A positive increase was observed in all mediums in the combination of 0.5 mg/l NAA or 0.5 mg/l 2. 4-D in the two cultivars used in this study. This study showed the genotypic difference effect on callus and bud formation.Roots were formed from allplantlets cultured on MS medium without plant hormones.Rooted plantlets were transferred into pots with nutrient soil, irrigated with water, and adapted tolaboratory conditions. Good plants grown to maturity were obtained in 12-13 weeks.
The method of vegetative propagation by hard wood cutting is considered of the most important ones of prevalent and successful in its cultivation to obtain a Punica Granatum seeding which are genetically uniform. In this research,it was used three categories fPunica Granatum ,Municipal,French,Darcoche,where they were taken the hard wood cutting of sound trees which free of insecticides and fungal infections and bacteria, and divided into groups which in cluded five deals to know the effect of hormone,stratification,impact,the hard wood cutting age and its length on rooting. It was found that the last three tested categories had no hormone effects on rooting. The soft wood cutting of one year-old overtook the ones of two-years of age,where they have the largest effect of rooting.The highest precentage of rooting and stronger were at the length of 30 cm. Its advised for stratification of the Municipal class befor its cultivation because it increases the rooting percentage. Darcoche is easy for rooting, because it roots by ahigh percentage if it had a stratification or impact by the conditions of continuous wetting before cultivation.
This present study was conducted to develop a detailed in vitro propagation system for the medicinal shrub Capparis spinosa L. Single nodes with one bud and a small part of stem of 1-1.5 cm long were used as initial explants which were collected f rom a shrubs grown under field conditions at Damascus suburb., (Doumar). Explants were surface-disinfected by 70% Ethanol for 1 min., followed by immersion in Sodium Hypochlorite or HgCl2 for different periods and concentrations with 1 drop of Tween 20 for 100 ml disinfectant solution, where after, they were placed onto MS basal medium containing a combination of growth regulators at different concentrations (BA at 4.44 or 8.88 μM) each with IBA 0.49 μM. Cultures were incubated in the growth room at 23±1 c and light intensity of 3000 lux at the cultures level. Multiplication rate of 25.17-fold from one explant was achieved every 4 weeks on the optimal MS medium (MS+8.88μM BA+0.49μM IBA). The described method has potential to produce large numbers of plantlets within a short period of time to expand its cultivation for medicinal uses.
This current study was carried out on B41 grape rootstock micropropagated in vitro at the General Commission for Scientific Agricultural Research (GCSAR), Syria to study the effect of different concentrations of sodium chloride on the multiplicati on and rooting stages. The results showed that the highest average of survival plants (98%), the number of new buds (8.43) and plant length (8.58 cm) were obtained in the control treatment after 4 weeks from multiplication stage. The treatment with 50 mM of sodium chloride led to reduce the multiplication rates with significant differences, while the lowest plant multiplication had occurred with 100 mM of sodium chloride and no survival plants were remained by the treatment with 150 mM of sodium chloride during multiplication stage. The highest rooting rates (% 85.30), the number of roots (4.67) and root length (6.28 cm) were also obtained in the control treatment after 4 weeks from rooting stage. The treatment with 50 mM of sodium chloride led to reduce the rooting process with significant differences. The lowest rooting results were observed with 100 mM of sodium chloride and no rooting rate was observed when plants were treated with 150 mM of sodium chloride.
