The use of engineered nanoscale magnetic materials in healthcare and biomedical technologies is rapidly growing. Two examples which have recently attracted significant attention are magnetic particle imaging (MPI) for biological monitoring, and magnetic field hyperthermia (MFH) for cancer therapy. Here for the first time, the capability of a Lissajous scanning MPI device to act as a standalone platform to support the application of MFH cancer treatment is presented. The platform is shown to offer functionalities for nanoparticle localization, focused hyperthermia therapy application, and non-invasive tissue thermometry in one device. Combined, these capabilities have the potential to significantly enhance the accuracy, effectiveness and safety of MFH therapy. Measurements of nanoparticle hyperthermia during protracted exposure to the MPI scanners 3D imaging field sequence revealed spatially focused heating, with a maximum that is significantly enhanced compared with a simple 1-dimensional sinusoidal excitation. The observed spatial heating behavior is qualitatively described based on a phenomenological model considering torques exerted in the Brownian regime. In-vitro cell studies using a human acute monocytic leukemia cell line (THP-1) demonstrated strong suppression of both structural integrity and metabolic activity within 24 h following a 40 min MFH treatment actuated within the Lissajous MPI scanner. Furthermore, reconstructed MPI images of the nanoparticles distributed among the cells, and the temperature-sensitivity of the MPI imaging signal obtained during treatment are demonstrated. In summary, combined Lissajous MPI and MFH technologies are presented; demonstrating for the first time their potential for cancer treatment with maximum effectiveness, and minimal collateral damage to surrounding tissues.

Visualization of the in vivo spatial distribution of superparamagnetic iron oxide nanoparticles (SPIONs) is crucial to biomedicine. Magnetic particle imaging (MPI) is one of the most promising approaches for direct measurements of the SPION distribution. In this paper, we systematically investigate a single-harmonic-based narrowband MPI approach. Herein, only the 3rd harmonic at 15 kHz of the SPION signal induced in an excitation magnetic field of 5 kHz is measured via a narrowband detection system for imaging during scanning a field-free-point in a field of view. Experiments on spot and line phantoms are performed to evaluate the spatial distribution by the assessment of the full width at half maximum and modulation transfer function at different excitation magnetic fields from 4 to 10 mT. Experimental results demonstrate that reconstructed images have a spatial resolution of 1.6 and 1.5 mm for a gradient field of 2.2 T/m and 4.4 T/m in x- and z-direction, respectively, at an excitation magnetic field of 4 mT. In terms of line gap, two lines with a gap of 0.5 mm are resolved. With increasing the excitation magnetic field to 10 mT, the spatial resolution gets worse to 2.4 and 2.0 mm in x- and z-direction, respectively. Moreover, the custom-built MPI scanner allows a limit of detection of 53 microgram (Fe)/mL (500 ng Fe weight) using perimag SPIONs. In addition, the excellent performance is demonstrated by imaging experiments on an emg logo phantom. We believe that the proposed narrowband MPI approach is a promising approach for SPION imaging.

Modern reconstruction methods for magnetic resonance imaging (MRI) exploit the spatially varying sensitivity profiles of receive-coil arrays as additional source of information. This allows to reduce the number of time-consuming Fourier-encoding steps by undersampling. The receive sensitivities are a priori unknown and influenced by geometry and electric properties of the (moving) subject. For optimal results, they need to be estimated jointly with the image from the same undersampled measurement data. Formulated as an inverse problem, this leads to a bilinear reconstruction problem related to multi-channel blind deconvolution. In this work, we will discuss some recently developed approaches for the solution of this problem.
Nanomagnetic hyperthermia (NMH) is intensively studied with the prospect of cancer therapy. A major challenge is to determine the dissipated power during in vivo conditions and conventional methods are either invasive or inaccurate. We present a non-calorimetric method which yields the heat absorbed during hyperthermia: it is based on accurately measuring the quality factor change of a resonant radio frequency circuit which is employed for the irradiation. The approach provides the absorbed power in real-time, without the need to monitor the sample temperature as a function of time. As such, it is free from the problems caused by the non-adiabatic heating conditions of the usual calorimetry. We validate the method by comparing the dissipated power with a conventional calorimetric measurement. We present the validation for two types of resonators with very different filling factors: a solenoid and a so-called birdcage coil. The latter is a volume coil, which is generally used in magnetic resonance imaging (MRI) under in vivo condition. The presented method therefore allows to effectively combine MRI and thermotherapy and is thus readily adaptable to existing imaging hardware.

Selective accumulation of B-10 compound in tumour tissue is a fundamental condition for the achievement of BNCT (Boron Neutron Capture Therapy), since the effectiveness of therapy irradiation derives just from neutron capture reaction of B-10. Hence, the determination of the B-10 concentration ratio, between tumour and healthy tissue, and a control of this ratio, during the therapy, are essential to optimise the effectiveness of the BNCT, which it is known to be based on the selective uptake of B-10 compound. In this work, experimental methods are proposed and evaluated for the determination in vivo of B-10 compound in biological samples, in particular based on neutron radiography and gammaray spectroscopy by telescopic system. Measures and Monte Carlo calculations have been performed to investigate the possibility of executing imaging of the 10B distribution, both by radiography with thermal neutrons, using 6LiF/ZnS:Ag scintillator screen and a CCD camera, and by spectroscopy, based on the revelation of gamma-ray reaction products from B-10 and the H. A rebuilding algorithm has been implemented. The present study has been done for the standard case of B-10 uptake, as well as for proposed case in which, to the same carrier, is also synthesized Gd-157, in the amount of is used like a contrast agent in NMRI.
Magnetic nanoparticle based hyperthermia emerged as a potential tool for treating malignant tumours. The efficiency of the method relies on the knowledge of magnetic properties of the samples; in particular, knowledge of the frequency dependent complex magnetic susceptibility is vital to optimize the irradiation conditions and to provide feedback for material science developments. We study the frequency-dependent magnetic susceptibility of an aqueous ferrite suspension for the first time using non-resonant and resonant radiofrequency reflectometry. We identify the optimal measurement conditions using a standard solenoid coil, which is capable of providing the complex magnetic susceptibility up to 150 MHz. The result matches those obtained from a radiofrequency resonator for a few discrete frequencies. The agreement between the two different methods validates our approach. Surprisingly, the dynamic magnetic susceptibility cannot be explained by an exponential magnetic relaxation behavior even when we consider a particle size-dependent distribution of the relaxation parameter.
