Coordination in circular and longitudinal muscle motions are of crucial importance in the motor function of gastrointestinal (GI) tract. Intestinal wall motions depend on myogenic-active properties of smooth muscles layers of intestinal wall, which is the ability to create active contractile forces in response to distension. Considering the stress in the circular and longitudinal smooth muscles as a sum of passive, depending on muscle deformations, and active, depending on muscle tone, components, and also assuming that the change in the muscle tone depends on the current stress-strain condition, the system of four ordinary differential equations (ODE) is obtained, which describes filling-emptying cycle of intestinal segment as a process of coordinated activities of circular and longitudinal muscles of intestinal wall. A general approach in formulating the modelling conditions is based on the previously described model restricted to the circularly distensible reservoir of constant length. Obtained results illustrate the character of coordinated activities of two orthogonal muscle layers, which are alternating phases of reciprocally and uniformly changing modalities such as stretching of the wall and muscle tone. The results also contribute to the existing understanding of the roles of Auerbachs and Meisners intermuscular and submucous neural plexuses in regulations of autonomous intestinal motility, as well as clarify functional mechanisms of the interstitial cells of Cajal (ICC) in triggering of smooth muscle contractions.

Ability of smooth muscles to contract in response to distension plays a crucial role in motor function of intestine. Qualitative analysis of dynamical models using myogenic active property of smooth muscles has shown well agreement with physiologic d ata. Considered as a self-regulatory unit, function of gastrointestinal (GI) segment is assumed to be regulated by integration of basis patterns providing accumulation and propagation of intestinal content. By implementing external, depending on neural system, variable to the previous model, and considering two attaches to one another reservoirs as a physical analogue of the segmental partition of intestine, a system of six ODE equations, three for each reservoir, describes coordinated wall motions and propagation of the content from one reservoir to another. It was shown that besides negative feedback (NFB), other functional patterns, namely positive feedback (PFB) and reciprocal links (RL) are involved in regulations of filling-emptying cycle. Being integrated in a whole functional system these three patterns expressed in a matrix form represent basis elements of imaginary part of coquaternion which with identity basis component is an algebraically closed structure under addition and multiplication of its elements. A coquaternion ring may be considered as a model of inner self-regulatory functional structure providing coordinated wall motions of GI tract portions.
We have developed a quantitative model for the creation of cytoplasmic Ca2+ gradients near the inner surface of the plasma membrane (PM). In particular we simulated the refilling of the sarcoplasmic reticulum (SR) via PM-SR junctions during asynchron ous [Ca2+] oscillations in smooth muscle cells of the rabbit inferior vena cava. We have combined confocal microscopy data on the [Ca2+] oscillations, force transduction data from cell contraction studies and electron microscopic images to build a basis for computational simulations that model the transport of calcium ions from Na+/Ca2+ exchangers (NCX) on the PM to sarcoplasmic/endoplasmic reticulum Ca2+ ATPase (SERCA) pumps on the SR as a three-dimensional random walk through the PM-SR junctional cytoplasmic spaces. Electron microscopic ultrastructural images of the smooth muscle cells were elaborated with software algorithms to produce a very clear and dimensionally accurate picture of the PM-SR junctions. From this study, we conclude that it is plausible and possible for enough Ca2+ to pass through the PM-SR junctions to replete the SR during the regenerative Ca2+ release, which underlies agonist induced asynchronous Ca2+ oscillations in vascular smooth muscle.
Some mechanisms of cardiac arrhythmias can be presented as a composition of elementary acts of block and reflection on the contacts of homogeneous areas of the conducting tissue. For study this phenomena we use an axiomatic one-dimensional model of i nteraction of cells of excitable tissue. The model has four functional parameters that determine the functional states durations of the cell. We show that the cells of a homogeneous excitable tissue, depending on the ratio of the durations of the functional intervals, can operate in the mode of solitary waves conduction or in one of three modes of selfgeneration. It is proved that the propagation of a solitary wave through the boundary of homogeneous conducting tissues can be accompanied by a block or multiplex reflection. Block and reflection are unidirectional phenomena, and there are not compatible on the same boundary. Systematized rules of transmitting, block and reflection waves at the boundary of homogeneous conducting tissues open up new possibilities for design mechanisms of generation and analyzing complex heart rate patterns.

As a first step in the search of an analytical study of mechanical denaturation of DNA in terms of the sequence, we study stable, stationary solutions in the discrete, finite and homogeneous Peyrard-Bishop DNA model. We find and classify all the stat ionary solutions of the model, as well as analytic approximations of them, both in the continuum and in the discrete limits. Our results explain the structure of the solutions reported by Theodorakopoulos {em et al.} [Phys. Rev. Lett. {bf 93}, 258101 (2004)] and provide a way to proceed to the analysis of the generalized version of the model incorporating the genetic information.
Rapidly dividing tissues, like intestinal crypts, are frequently chosen to investigate the process of tumor initiation, because of their high rate of mutations. To study the interplay between normal and mutant as well as immortal cells in the human c olon or intestinal crypt, we developed a 4-compartmental stochastic model for cell dynamics based on current discoveries. Recent studies of the intestinal crypt have revealed the existence of two stem cell groups. Therefore, our model incorporates two stem cell groups (central stem cells (CeSCs) and border stem cells (BSCs)), plus one compartment for transit amplifying (TA) cells and one compartment of fully differentiated (FD) cells. However, it can be easily modified to have only one stem cell group. We find that the worst-case scenario occurs when CeSCs are mutated, or an immortal cell arises in the TA or FD compartments. The probability that the progeny of a single advantageous CeSC mutant will take over the entire crypt is more than $0.2$, and one immortal cell always causes all FD cells to become immortals.Moreover, when CeSCs are either mutants or wild-type (w.t.) individuals, their progeny will take over the entire crypt in less than 100 days if there is no immortal cell. Unexpectedly, if the CeSCs are wild-type, then non-immortal mutants with higher fitness are washed out faster than those with lower fitness. Therefore, we suggest one potential treatment for colon cancer might be replacing or altering the CeSCs with the normal stem cells.
