In this work we consider the inverse problem of reconstructing the optical properties of a layered medium from an elastography measurement where optical coherence tomography is used as the imaging method. We hereby model the sample as a linear dielectric medium so that the imaging parameter is given by its electric susceptibility, which is a frequency- and depth-dependent parameter. Additionally to the layered structure (assumed to be valid at least in the small illuminated region), we allow for small scatterers which we consider to be randomly distributed, a situation which seems more realistic compared to purely homogeneous layers. We then show that a unique reconstruction of the susceptibility of the medium (after averaging over the small scatterers) can be achieved from optical coherence tomography measurements for different compression states of the medium.

In this paper, we consider visualization of displacement fields via optical flow methods in elastographic experiments consisting of a static compression of a sample. We propose an elastographic optical flow method (EOFM) which takes into account expe rimental constraints, such as appropriate boundary conditions, the use of speckle information, as well as the inclusion of structural information derived from knowledge of the background material. We present numerical results based on both simulated and experimental data from an elastography experiment in order to demonstrate the relevance of our proposed approach.
The inverse problem we consider is to reconstruct the location and shape of buried obstacles in the lower half-space of an unbounded two-layered medium in two dimensions from phaseless far-field data. A main difficulty of this problem is that the tra nslation invariance property of the modulus of the far field pattern is unavoidable, which is similar to the homogenous background medium case. Based on the idea of using superpositions of two plane waves with different directions as the incident fields, we first develop a direct imaging method to locate the position of small anomalies and give a theoretical analysis of the algorithm. Then a recursive Newton-type iteration algorithm in frequencies is proposed to reconstruct extended obstacles. Finally, numerical experiments are presented to illustrate the feasibility of our algorithms.

This article is concerned with the derivation of numerical reconstruction schemes for the inverse moving source problem on determining source profiles in (time-fractional) evolution equations. As a continuation of the theoretical result on the unique ness, we adopt a minimization procedure with regularization to construct iterative thresholding schemes for the reduced backward problems on recovering one or two unknown initial value(s). Moreover, an elliptic approach is proposed to solve a convection equation in the case of two profiles.
Optical tweezers are an invaluable tool for non-contact trapping and micro-manipulation, but their ability to facilitate high-throughput volumetric microrheology of biological samples for mechanobiology research is limited by the precise alignment as sociated with the excitation and detection of individual bead oscillations. In contrast, radiation pressure from a low numerical aperture optical beam can apply transversely localized force over an extended depth range. We propose photonic force optical coherence elastography (PF-OCE), leveraging phase-sensitive interferometric detection to track sub-nanometre oscillations of beads, embedded in viscoelastic hydrogels, induced by modulated radiation pressure. Since the displacements caused by ultra-low radiation-pressure force are typically obscured by absorption-mediated thermal effects, mechanical responses of the beads were isolated after independent measurement and decoupling of the photothermal response of the hydrogels. Volumetric imaging of bead mechanical responses in hydrogels with different agarose concentrations by PF-OCE was consistent with bulk mechanical characterization of the hydrogels by shear rheometry.
In this paper, we consider the problem of estimating the internal displacement field of an object which is being subjected to a deformation, from Optical Coherence Tomography (OCT) images before and after compression. For the estimation of the intern al displacement field we propose a novel algorithm, which utilizes particular speckle information to enhance the quality of the motion estimation. We present numerical results based on both simulated and experimental data in order to demonstrate the usefulness of our approach, in particular when applied for quantitative elastography, when the material parameters are estimated in a second step based on the internal displacement field.
