An important goal of environmental health research is to assess the risk posed by mixtures of environmental exposures. Two popular classes of models for mixtures analyses are response-surface methods and exposure-index methods. Response-surface methods estimate high-dimensional surfaces and are thus highly flexible but difficult to interpret. In contrast, exposure-index methods decompose coefficients from a linear model into an overall mixture effect and individual index weights; these models yield easily interpretable effect estimates and efficient inferences when model assumptions hold, but, like most parsimonious models, incur bias when these assumptions do not hold. In this paper we propose a Bayesian multiple index model framework that combines the strengths of each, allowing for non-linear and non-additive relationships between exposure indices and a health outcome, while reducing the dimensionality of the exposure vector and estimating index weights with variable selection. This framework contains response-surface and exposure-index models as special cases, thereby unifying the two analysis strategies. This unification increases the range of models possible for analyzing environmental mixtures and health, allowing one to select an appropriate analysis from a spectrum of models varying in flexibility and interpretability. In an analysis of the association between telomere length and 18 organic pollutants in the National Health and Nutrition Examination Survey (NHANES), the proposed approach fits the data as well as more complex response-surface methods and yields more interpretable results.

The use of a finite mixture of normal distributions in model-based clustering allows to capture non-Gaussian data clusters. However, identifying the clusters from the normal components is challenging and in general either achieved by imposing constraints on the model or by using post-processing procedures. Within the Bayesian framework we propose a different approach based on sparse finite mixtures to achieve identifiability. We specify a hierarchical prior where the hyperparameters are carefully selected such that they are reflective of the cluster structure aimed at. In addition this prior allows to estimate the model using standard MCMC sampling methods. In combination with a post-processing approach which resolves the label switching issue and results in an identified model, our approach allows to simultaneously (1) determine the number of clusters, (2) flexibly approximate the cluster distributions in a semi-parametric way using finite mixtures of normals and (3) identify cluster-specific parameters and classify observations. The proposed approach is illustrated in two simulation studies and on benchmark data sets.

Quantile regression models are a powerful tool for studying different points of the conditional distribution of univariate response variables. Their multivariate counterpart extension though is not straightforward, starting with the definition of multivariate quantiles. We propose here a flexible Bayesian quantile regression model when the response variable is multivariate, where we are able to define a structured additive framework for all predictor variables. We build on previous ideas considering a directional approach to define the quantiles of a response variable with multiple-outputs and we define noncrossing quantiles in every directional quantile model. We define a Markov Chain Monte Carlo (MCMC) procedure for model estimation, where the noncrossing property is obtained considering a Gaussian process design to model the correlation between several quantile regression models. We illustrate the results of these models using two data sets: one on dimensions of inequality in the population, such as income and health; the second on scores of students in the Brazilian High School National Exam, considering three dimensions for the response variable.
Environmental health studies are increasingly measuring multiple pollutants to characterize the joint health effects attributable to exposure mixtures. However, the underlying dose-response relationship between toxicants and health outcomes of interest may be highly nonlinear, with possible nonlinear interaction effects. Existing penalized regression methods that account for exposure interactions either cannot accommodate nonlinear interactions while maintaining strong heredity or are computationally unstable in applications with limited sample size. In this paper, we propose a general shrinkage and selection framework to identify noteworthy nonlinear main and interaction effects among a set of exposures. We design hierarchical integrative group LASSO (HiGLASSO) to (a) impose strong heredity constraints on two-way interaction effects (hierarchical), (b) incorporate adaptive weights without necessitating initial coefficient estimates (integrative), and (c) induce sparsity for variable selection while respecting group structure (group LASSO). We prove sparsistency of the proposed method and apply HiGLASSO to an environmental toxicants dataset from the LIFECODES birth cohort, where the investigators are interested in understanding the joint effects of 21 urinary toxicant biomarkers on urinary 8-isoprostane, a measure of oxidative stress. This work relates the framework of model-based clustering for spatial functional data where the data are surfaces. We first introduce a Bayesian spatial spline regression model with mixed-effects (BSSR) for modeling spatial function data. The BSSR model is based on Nodal basis functions for spatial regression and accommodates both common mean behavior for the data through a fixed-effects part, and variability inter-individuals thanks to a random-effects part. Then, in order to model populations of spatial functional data issued from heterogeneous groups, we integrate the BSSR model into a mixture framework. The resulting model is a Bayesian mixture of spatial spline regressions with mixed-effects (BMSSR) used for density estimation and model-based surface clustering. The models, through their Bayesian formulation, allow to integrate possible prior knowledge on the data structure and constitute a good alternative to recent mixture of spatial spline regressions model estimated in a maximum likelihood framework via the expectation-maximization (EM) algorithm. The Bayesian model inference is performed by Markov Chain Monte Carlo (MCMC) sampling. We derive two Gibbs sampler to infer the BSSR and the BMSSR models and apply them on simulated surfaces and a real problem of handwritten digit recognition using the MNIST data set. The obtained results highlight the potential benefit of the proposed Bayesian approaches for modeling surfaces possibly dispersed in particular in clusters.

In the era of big data, variable selection is a key technology for handling high-dimensional problems with a small sample size but a large number of covariables. Different variable selection methods were proposed for different models, such as linear model, logistic model and generalized linear model. However, fewer works focused on variable selection for single index models, especially, for single index logistic model, due to the difficulty arose from the unknown link function and the slow mixing rate of MCMC algorithm for traditional logistic model. In this paper, we proposed a Bayesian variable selection procedure for single index logistic model by taking the advantage of Gaussian process and data augmentation. Numerical results from simulations and real data analysis show the advantage of our method over the state of arts.
